Western Blot Validation in Mouse A-20 Cell Lines. Loading: 15 µg of lysates per lane. Antibodies: TSLP orb1240269 (A: 0.5 µg/mL, B: 1 µg/mL, C: 2 µg/mL), 1h incubation at RT in 5% NFDM/TBST. Secondary: Goat anti-rabbit IgG HRP conjugate at 1:10000 dilution.

Immunofluorescence Validation of TSLP in Mouse Brain Tissue. Immunofluorescent analysis of 4% paraformaldehyde-fixed Mouse Brain Tissue labeling TSLP with orb1240269 at 20 µg/mL, followed by goat anti-rabbit IgG secondary antibody at 1/500 dilution (red).

Immunohistochemistry Validation of TSLP in Mouse Brain Tissue. Immunohistochemical analysis of paraffin-embedded Mouse Brain Tissue using anti-TSLP antibody (orb1240269) at 2.5 µg/ml. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.

Induction Validation of TSLP in Nasal Mucosa of Allergic Rhinitis (AR) mice (Wang et al., 2012). Western Blot images showing theTSLP protein detected by anti-mouse TSLP antibodies in normal mice (Control) or AR mice. TSLP protein expression levels were higher in AR mice and AR-GFP mice.

Induction Validation of TSLP in Rat VSMCs (Zhao et al., 2012). Immunohistochemical images showing the TSLP protein detected by anti-TSLP antibodies in Rat VSMCs without (Control) or with treatment with TNF-alpha, Ang II, Ang II+PDTC, Ang II +valsartan. TNF-α treatment is used as a positive control. The production of TSLP was increased after Ang II treatment.
