Application notes | Neutralization:To yield one-half maximal inhibition [ND50] of the biological activity of hTECK (100 ng/mL), a concentration of 10.0-20.0 μg/mL of this antibody is required.ELISA:In a sandwich ELISA (assuming 100 μL/well), a concentration of 1.0-2.0 μg/mL of this antibody will detect at least 300 pg/mL of recombinant human TECK when used with our biotinylated antigen affinity purified anti-human TECK (XP-5280Bt) as the detection antibody at a concentration of approximately 0.5-1.0 μg/mL. Western Blot:To detect hTECK by Western Blot analysis this antibody can be used at a concentration of 0.25-0.50 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant hTECK is 0.50-1.0 ng/lane, under non-reducing conditions. |

This antibody stained formalin-fixed, paraffin-embedded sections of human normal small bowel. The recommended concentration is 2.5 ug/ml with an overnight incubation at 4°C. An HRP-labeled polymer detection system was used with a DAB chromogen and a Proteinase K antigen retrieval. Optimal concentrations and conditions may vary.
