  Further information: Immunohistochemistry: This was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration was determined to be 10 ug/ml.

IF (ICC) notes: ICC/IF analysis of CD73 in HeLa cells. The cell was stained with CD73 antibody (1:100). The secondary antibody (green) was used Alexa Fluor 488. DAPI was stained the cell nucleus (blue).

Cell lysates of human mesenchymal stem cell (30 ug) were resolved by SDS-PAGE, transferred to NC membrane and probed with anti-human CD73 (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL.

The cell lysates (40ug) were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human CD73 antibody (1:1000).
