• Dilution range: Peptide ELISA: antibody detection limit dilution 1:16000. Western blot: Approx. 60kDa band observed in lysates of cell lines Daudi and U937, and in nuclear cell lysates of HEK293 (calculated MW of 53.0kDa according to NP_001243481.1). This band was successfully blocked by incubation with the immunizing peptide. Recommended concentration: 2-3µg/ml. Primary incubation 1 hour at room temperature. IHC: In paraffin embedded Human Placenta shows strong staining of cytoplasm in syncytiotrophoblasts. Recommended concentration: 5-10µg/ml. Immunofluorescence: Strong expression of the protein seen in the nuclei and cytoplasm of U937 cells. Recommended concentration: 10µg/ml. Flow Cytometry: Flow cytometric analysis of U937 cells. Recommended concentration: 10ug/ml.

Immunofluorescence analysis of paraformaldehyde fixed U937 immobilized on Shi-fix™ plus cover-slips. Primary incubation 1hr (1:50 dilution) followed by Alexa Fluor® 488 secondary antibody (1:2000 dilution), showing cytoplasmic and nuclear staining. The nuclear stain is DAPI (blue). Negative control: Anti-Goat IgG followed by Alexa Fluor® 488 secondary antibody.

Flow cytometric analysis of paraformaldehyde fixed U937 cells (blue line), permeabilized with 0.5% Triton. Primary incubation 1hr (10 ug/ml) followed by Alexa Fluor 488 secondary antibody (1 ug/ml). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody.

5 µg/ml staining of paraffin embedded Human Placenta. Steamed antigen retrieval with citrate buffer pH6, AP-staining.

Primary incubation 1 hour at room temperature. Images A+B: Daudi cell lysate + peptide, C+D: U937 cell lysate + peptide, E+F: HEK293 nuclear cell lysate + peptide, incubation at primary Ab concentration 3 ug/ml. (Loaded 35 µg protein in RIPA buffer, per lane). Detected by chemiluminescence.
