CategoryAntibodies
DescriptionEstrogen receptor (ER) is an important regulator of growth and differentiation in the mammary gland. ER presence in breast tumors indicates an increased likelihood of response to anti-estrogen (e.g. tamoxifen) therapy. Clone ER506 antibody strongly stains nuclei of epithelial cells in breast carcinomas.
ClonalityMonoclonal
Species/HostMouse
IsotypeMouse IgG1, kappa
ConjugationUnconjugated
ReactivityHuman
ImmunogenRecombinant human ER alpha protein (aa 2-185) was used as the immunogen.
Tested applicationsFACS, IHC-P, WB
Dilution rangeFlow cytometry: 1-2ug/million cells,Western blot: 1-2ug/ml,Immunohistochemistry (FFPE): 1-2ug/ml (1)
Application notesVariations in protocols, secondaries and substrates may require the ER antibody to be titered for optimal performance.1. FFPE testing requires boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 10-20 min and cooling at RT for 20 minutes.
Antibody TypePrimary Antibody
Clone NumberER506
Formula0.2 mg/ml in 1X PBS with 0.1 mg/ml BSA (US sourced) and 0.05% sodium azide
StorageMaintain refrigerated at 2-8°C for up to 2 weeks. For long term storage store at -20°C in small aliquots to prevent freeze-thaw cycles.
Hazard InformationThis ER antibody is available for research use only.

Estrogen Receptor alpha Antibody

IHC testing of FFPE human endometrium with ER antibody (clone ER506).

Estrogen Receptor alpha Antibody

Intracellular FACS testing of human MCF-7 cells: Black = cells alone; Green = isotype control; Red = PE conjugated ER antibody (V2113PE)

Estrogen Receptor alpha Antibody

Western blot testing of MCF-7 cell lysate with ER antibody (clone ER506). Predicted molecular weight of ER alpha: ~67 kDa.

Estrogen Receptor alpha Antibody

Analysis of HuProt (TM) microarray containing more than 19, 000 full-length human proteins using ER antibody (clone ER506). These results demonstrate the foremost specificity of the ER506 mAb. Z- and S- score: The Z-score represents the strength of a signal that an antibody (in combination with a fluorescently-tagged anti-IgG secondary Ab) produces when binding to a particular protein on the HuProt (TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If the targets on the HuProt (TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-scores. The S-score therefore represents the relative target specificity of an Ab to its intended target.
