Astrocytes (1 × 106 cells/ml) were cultured for 24 h without treatment or with METH (10 μM), clade B gp120 (50 ng/ ml), or clade C gp120 (50 ng/ ml) alone or METH in combination with clade B or C gp120. After incubation, the cells were fixed, permeabilized, and stained for (A) DRD-2, (B) phospho-CaMK II (Cat. orb319474) (C) phospho-CaMK IV (green) or GFPA (red) and DAPI (blue). The cells were imaged using a confocal microscope at 630x and analyzed using immunocytochemistry.

Depicts increased CaMKII autophosphorylation at Threonine 287 (Cat. orb319474, 1:1000) (normalized to CaMKIIδ expression) following ATX-II exposure in WT and KI cells.

Representative immunoblot and quantification of in vitro kinase reactions at 30 °C in 1 mM ATP measuring CaMKIIα WT and F89G phosphorylation of T286 (Cat. orb319474, 1:2500).

Representative immunoblot and quantification of in vitro kinase reactions at 30 °C in 1 mM ATP measuring AS283-mediated inhibition of CaMKII phosphorylation of GluA1 S831 (pS831) (Cat. orb319552, 1:2000) and autophosphorylation of CaMKII T286 (pT286) (Cat. orb319474, 1:2500) with varying concentrations of AS283 (0.41–10 μM). For a negative control (Neg.), ATP was omitted.

Western blot of rat brain lysate showing specific immunolabeling of the ~50 kDa α- and the ~60 kDa β-CaM Kinase II phosphorylated at Thr286 in the first lane (-). Phosphospecificity is shown in the second lane (+) where the immunolabeling is completely eliminated by blot treatment with lambda phosphatase (λ-Ptase, 1200 units for 30 minutes).

Western immunoblot measuring levels of T286 phosphorylation of CaMKII (Cat. orb319474,1:1000) normalized to total CaMKII in the membrane fraction of whole hippocampi from sham and CA/CPR operated mice.
