Description

Hypericin, also known as SGX301, is a novel, first-in-class photodynamic therapy using a potent photosensitizer, synthetic hypericin, that is topically applied and activated by safe visible fluorescent light. This treatment avoids the risk of secondary malignancies (including melanoma) inherent with the frequently employed DNA-damaging chemotherapeutic drugs and other photodynamic therapies that are dependent on ultraviolet A (UVA) light exposure.

Bioactivity

In Vivo
Animal model: 18-20 week-old female BALB/c mice injected withCT26 carcinomas. Dosage: 10 mg/kg. Administration: Intravenous injection; 10 mg/kg; once. Result: Showed a four times delayed tumor growth compared to the control groups.
In Vitro
Cell Viability Assay Cell line: Fibroblasts (Fb), melanocytes (Mc), and keratinocytes (Kc). Concentration: 0.25 μM; 0.5 μM; 1 μM; 2 μM; 3 μM; 4 μM Incubation time: 24 hours. Result: Showed the LD50 for Fb and Mc at 1.75 μM and 3.5 μM, respectively, and for Kc at a greater dose than 4 μM. Apoptosis Analysis Cell line: Fibroblasts (Fb), melanocytes (Mc), and keratinocytes (Kc). Concentration: 3 μM. Incubation time: 24 hours. Result: Showed a significant (p<0.001) early apoptotic Fb population (64%), and a smaller, significant (p<0.05) early apoptotic Mc population (20%).
