  Anti-Choline Oxidase has been tested by ELISA and western blot. This product is suitable to be assayed against 1.0 ug of Choline Oxidase in a standard capture ELISA using Peroxidase Conjugated Streptavidin and ABTS (2,2’-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) code as a substrate for 30 minutes at room temperature. A working dilution of 1:4,000 to 1:20,000 of the reconstitution concentration is suggested for this product. Reconstitution Buffer: Restore with deionized water (or equivalent). Reconstitution Volume: 100 µL

Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Biotin, anti-Goat Serum as well as purified and partially purified Choline Oxidase [Alcaligenes species]. Cross reactivity against Choline Oxidase from other sources is unknown.

• Storage: Store vial at 4° C prior to restoration. For extended storage aliquot contents and freeze at -20° C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.

Biorbyt Goat anti Choline Oxidase antibody was used to detect Choline Oxidase. Samples of ~1 and 0.25 ug of purified Choline Oxidase per lane were run by SDS-PAGE under reducing and non-reducing conditions and reduced samples of purified Choline Oxidase contained 4% BME and were boiled for 5 minutes. Protein was transferred to nitrocellulose and probed with Biorbyt Goat anti Choline Oxidase (ON 4 C). Primary antibody was detected with Biorbyt Dylight 649 conjugated Donkey anti Goat (1:10K 1.5 hr RT in orb348637).

Primary antibody: Goat anti-Choline Oxidase (Alcaligenes species) Antibody Biotin Conjugated at 1:1000 overnight at 4°C. Secondary antibody: HRP streptavidin secondary antibody at 1:40000 for 30 min at RT. Block: orb348637 for 30 min at RT. Predicted/Observed size: 60 kDa, 60 kDa for Choline Oxidase.
