  Anti-Uricase Antibody has been tested by ELISA and western blot. This product is suitable to be assayed against 1.0 ug of Uricase in a standard capture ELISA using Peroxidase Conjugated Streptavidin #S000-03 and ABTS (2,2’-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:4,000 to 1:53,000 of the reconstitution concentration is suggested for Anti-Uricase Antibody.

• Purity: Anti-Uricase Antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Biotin, anti-Goat Serum as well as purified and partially purified Uricase [Bacillus species]. Cross reactivity against Uricase from other sources is unknown.

Biorbyt Goat anti Uricase antibody was used to detect purified Uricase under reducing and non-reducing conditions. Samples of ~1 and 0.25 ug of protein per lane were run by SDS-PAGE and reduced samples of purified Uricase contained 4% BME and were boiled for 5 minutes. Protein was transferred to nitrocellulose and probed with Biorbyt Goat anti Uricase. Primary antibody was detected with Biorbyt Dylight 649 conjugated Donkey anti Goat (1:10K 1.5 hr RT in orb348637).
