Flow cytometry using the Anti-S100A9 antibody IID4B10. Paraformaldehyde fixed U937 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (orb256449; isotype control, black line) or the mouse IgG1 version of IID4B10 (orb1821617, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Immunofluorescence staining of U937 cells with Anti-S100A9 IID4B10. Immunofluorescence analysis of paraformaldehyde fixed U937 cells immobilized on Shi-fix™ plus cover-slips, permeabilized with 0.15% Triton stained with the chimeric mouse IgG1 version of IID4B10 (orb1821617) (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing nuclear and cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom orb1821617, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (orb256449) followed by staining with Alexa Fluor® 488 secondary antibody.

Western Blot using Anti-S100A9 antibody IID4B10. Human PBM(A) (0.01 µg/ml), human spleen(B) (0.1 ug/ml), human tonsil(C) (0.001 ug/ml) and human bone marrow(D) (0.001 ug/ml) tissue lysates (35 µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric mouse version of IID4B10 (orb1821617) before detection using an anti-mouse secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
