Flow-cytometry using the anti-PD-1 antibody 29F.1A12. Mouse splenocytes were stained with an isotype control (orb256390, black line) or the rabbit-chimeric version of 29F.1A12 (orb2645605, blue line) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using Alexa Fluor 488® conjugated goat anti-rabbit IgG and cells analysed on a FACSCanto flow-cytometer.

Western Blot using anti-PD-1 antibody 29F.1A12. Mouse thymus (1 µg/ml) lysate (35 µg protein in RIPA buffer) was resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of X77 (orb2645605) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
