Flow cytrometry using anti-IL-18 antibody ABT-325. Human blood leucocytes were rested for 4h at 37°C, fixed with 2% PFA, permeabilized with 0.5% Triton, and stained with the anti-unknown specificity antibody (orb256458; negative control, right) or the rabbit IgG version of ABT-325 (orb2645792, left) at a dilution of 1:100 overnight at 4°C. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.

Western blot using anti-IL-18 antibody ABT-325. HeLa cells lysate (35 µg protein in RIPA buffer) was resolved via SDS-PAGE, and the subsequent blot was probed with the chimeric rabbit version of ABT-325 (orb2645792) at 1 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1 hour was used, and proteins were detected by chemiluminescence.
