Flow cytometry using the Anti-HDGF antibody H3. Paraformaldehyde fixed MCF7 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (orb256449; isotype control, black line) or the mouse IgG1 version of H3 (orb1821619, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Immunofluorescence staining of MCF7 cells with Anti-HDGF H3. Immunofluorescence analysis of paraformaldehyde fixed MCF7 cells permeabilized with 0.15% Triton stained with the chimeric mouse IgG1 version of H3 (orb1821619) (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom orb1821619, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (orb256449) followed by staining with Alexa Fluor® 488 secondary antibody.

Western Blot using Anti-HDGF antibody H3. HeLa(A) (0.0001 ug/ml), HeLa nuclear (B) (0.0001 ug/ml), MCF7(C) (0.0001 ug/ml), MCF7 nuclear (D) (0.0001 ug/ml), HepG2(E) (0.0001 ug/ml), HepG2 nuclear(F) (0.0001 ug/ml), A549(G) (0.0001 ug/ml) and A431(H) (0.0001 ug/ml) cell lysate (35 µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric mouse version of H3 (orb1821619) 0.001 µg/ml before detection using an anti-mouse secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
