ImmunogenThis antibody was raised by immunising Wistar rats with the CD25+CD4+ T cell line. Hybridoma cells were generated by fusing spleen cells from the immunized rat with P3X63Ag8.653 myeloma cells, assessed for secreting antibodies capable of neutralizing in vitro CD25+CD4+ T cell-mediated suppression of anti-CD3-stimulated T cell proliferation9, and then cloned. This DTA-1 clone secreted a monoclonal rat immunoglobulin G2a (IgG2a) that showed high neutralizing activity.

Anti-GITR [DTA-1]

Flow cytometry using the Anti-GITR antibody DTA-1. Paraformaldehyde fixed mouse splenocytes were stained with anti-unknown specificity antibody (orb256458; isotype control, black line) or the rabbit IgG version of DTA-1 (orb758819, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Immunofluorescence staining of fixed mouse splenocytes with anti-GITR antibody DTA-1. Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes on Shi-fix™ coverslips stained with the chimeric rabbit IgG version of DTA-1 (orb758819) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom orb758819, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (orb256449) followed by staining with Alexa Fluor® 488 secondary antibody.
