VVD-484 is a PI3Kp110α inhibitor with an IC50 of 0.59 μM for human targets. Classified as a "silent ligand," VVD-484 covalently binds to cysteine 242 of PI3Kp110α without disrupting the p110α-KRASG12C interaction. It inhibits AKT phosphorylation (S473) through a RAS-independent pathway. VVD-484 is applicable in the study of HER2 overexpressing cancers.
