Description

SY-589 is an orally active inhibitor of the DNA polymerase Polθ helicase domain (IC50=2.29 nM) and an inducer of DNA damage. It inhibits the ATPase activity of the Polθ helicase domain, blocking the Polθ-mediated microhomology-mediated end-joining (MMEJ) DNA repair pathway (IC50=0.85 nM). Additionally, SY-589 induces the accumulation of DNA double-strand breaks by increasing γ-H2AX levels. This compound exhibits antiproliferative effects on BRCA2-deficient cells and is useful for studying tumors with homologous recombination (HR) deficiencies.
