Ro 67-4853 is a selective positive allosteric modulator of metabotropic glutamate receptor 1 (mGluR1). It potentiates agonist responses, such as to DHPG, and is used in neuroscience research to study mGluR1 function in both in vitro assays and in vivo models, including iontophoretic studies in anesthetized rats.

• In Vitro:
  Ro 67-4853 (1 μM) was preincubated with BHK cells stably expressing mGluR1a for 10 min before the addition of a range of glutamate concentrations. These compounds had no agonist activity at mGluR1 when added alone, but the concentration-response curve for glutamate was shifted to the left approximately 15-fold.
