Bioactivity

• In Vivo:
  METHODS: A single oral dose of PQR514 (10 mg/kg) was administered to male C57BL/6J mice at plasma concentrations of 4 and brain and thigh muscle concentrations were monitored over time. RESULTS: PQR514(4) reached maximum concentrations in plasma (3282ng/mL) after 1 hour and in brain and thigh muscles after 30 minutes (1023 and 1808 ng/g, respectively). METHODS: Using the OVCAR-3 xenograft model in BALB/c nude mice, the treatment group received PQR514 (6.3 mg/kg, p.o. QD 28 days) starting on day 17 to evaluate the in vivo anti-proliferative effect of PQR514. RESULTS: PQR514 treatment resulted in significant anti-tumor activity, with PQR514 significantly inhibiting tumor growth compared to control mice.
