Background | PGLa is an antibiotic peptide member of the magainin family, originally identified in the skin of the African frog Xenopus laevis and its secretions. PGLa has antibacterial, antifungal, and antiviral properties, acting on gram-negative and gram-positive bacteria via disruption of the cellular lipid bilayer and inducing transmembrane pore formation. In common with other magainins, PGLa does not exhibit hemolytic activity. When used in combination with magainin 2, PGLa has a synergistic effect, which it also demonstrates when used in combination with other antibiotics. PGLa can also restore antibiotic activity in resistant bacteria, and inhibit evolution of antibiotic resistance. Used in combination, a sub-inhibitory dose of PGLa causes a 30 fold increase in susceptibility in antibiotic resistant bacteria, and the same sub-inhibitory dose efficiently slows down the development of antibiotic resistance. |
