Description

Pap12-6-10, an MD-2 ligand, binds to the MD-2 hydrophobic pocket to inhibit the dimerization of the TLR4/MD-2 complex and downstream inflammatory signaling. It can also penetrate bacterial cell membranes via LPS, inducing oxidative stress and causing bacterial death. Pap12-6-10 regulates the LPS-induced inflammatory response through the TLR4 signaling pathway and exhibits antimicrobial activity against multidrug-resistant Gram-negative bacteria. It shows a low propensity for resistance development and exhibits minimal cytotoxicity in preclinical studies, additionally preventing organ damage in septic mouse models. Pap12-6-10 is applicable in research related to Gram-negative sepsis and carbapenem-resistant Acinetobacter baumannii infections.
