Neriifolin is a cardiac glycoside and Na+/K+-ATPase inhibitor derived from Cerbera manghas. It exhibits anticancer properties by inducing cell cycle arrest and apoptosis in vitro in leukemia (THP-1) and liver cancer (HepG2) cell lines, and is also used in prostate cancer research.

Bioactivity

• In Vitro:
  Treatment of Neriifolin at a concentration of 0.1 μg/mL for 48 hours was able to cause apoptosis of HepG2 cells. In addition, Neriifolin significantly reduced the viability of HepG2 cells after 72 hours of treatment at concentrations ranging from 0 to 8 μg/mL. The compound also caused cell cycle arrest in the S and G2/M phases, further promoting apoptosis in HepG2 cells. When HepG2 cells were stimulated by Neriifolin, caspase-3, caspase-8, and caspase-9 were activated, while the expression of Fas and FasL proteins was upregulated.
