Description

MNP2 is an inhibitor of the NLRP3-ASC interaction. It selectively binds to the PYD domain of ASC (Ka=149 nM), blocking the ASC-PYM binding (Ka=58 nM), which subsequently inhibits the interaction between ASC and NLRP3, and prevents the formation of the NLRP3 inflammasome. MNP2 effectively suppresses the release of IL-1β and the maturation of caspase-1, and reduces the efflux of potassium and chloride ions. It also prevents mitochondrial damage and the production of reactive oxygen species, significantly reducing the formation of the NLRP3 inflammasome induced by β-amyloid, Tau protein, and α-synuclein in neurodegenerative lesions. MNP2 has potential applications in the study of neurodegenerative diseases.
