Ezurpimtrostat hydrochloride (GNS561 hydrochloride) is an orally bioavailable, small-molecule inhibitor of palmitoyl-protein thioesterase 1 (PPT1). It exerts anticancer and antifibrotic activity by blocking late-stage autophagy, leading to lysosomal dysfunction and the inhibition of cancer cell proliferation, as demonstrated in in vitro and in vivo studies.
