Description

DIM-3,5-Cl2 is an inverse agonist of NR4A1 (KD = 7.7 μM) and NR4A2 (KD = 12.0 μM) and possesses bidirectional regulatory functions. As an inverse agonist, DIM-3,5-Cl2 suppresses the expression of targets that promote cancer and endometriosis; as an agonist, it enhances NR4A1/2-mediated transcriptional activation of CD71 in cooperation with Sp1/Sp4. DIM-3,5-Cl2 induces tumor cell death through two lethal signaling pathways: one relies on elevated ROS, exacerbated lipid peroxidation, and MDA accumulation, along with inhibition of GPX4/SLC7A11 to trigger ferroptosis; the other induces apoptosis via cleavage-mediated activation of PARP/caspase-3 and downregulation of BCL-2.
