Peroxidase chromogen-substrate 3-3´ diaminobezidine (DAB) reagent is an excellent chromogen system that is used in Immunohistochemistry (IHC) and Immunocytochemistry (ICC).

• Application Notes:
  To prepare the reagent, add two drops of Chromogen reagent to one milliliter of Buffer Substrate in a test tube and mix well. This ready-to-use reagent remains stable for several hours, and any unused DAB solution should be discarded according to local regulations. For IHC and ICC applications, following the peroxidase reaction, wash the slide with buffer and distilled or deionized water. Apply a few drops of the ready-to-use DAB reagent to the tissue slides for 5 to 10 minutes at room temperature. Wash two to five times with buffer, followed by a rinse with distilled or deionized water. Apply a counterstain compatible with DAB, such as hematoxylin, for 30 to 60 seconds. Wash with tap water or a buffer with a pH of 7.4 or higher, followed by a rinse with distilled or deionized water. Finally, mount the slide with aqueous or organic mounting medium after dehydration as specified in the mounting protocol.
