CUR5g is a small molecule autophagy inhibitor that suppresses A549 cell migration and colony formation by blocking STX17 recruitment to autophagosomes via a UVRAG-dependent mechanism. It is widely used in cancer research, particularly in combination with cisplatin, for in vitro studies of autophagy's role in tumor progression and chemoresistance.

Bioactivity

• In Vivo:
  CUR5g (40 mg/kg) and cisplatin (1 mg/kg), administered via the caudal vein every other day for a total of 15 days, were able to retard the growth of xenografted tumors. The combined treatment of CUR5g and cisplatin nearly completely inhibited tumor growth.

• In Vitro:
  In A549 cells, CUR5g (at concentrations of 0, 1, 5, 10, 20, and 40 μM; observed at 3, 6, 12, and 24 hours) induces extensive cytoplasmic vacuolization, and the GFP-LC3B signal shifts from diffuse cytosolic staining to a punctate pattern outlining autophagosomes.
