Description

GDC-4198 (Compound EX.161) is an orally active, blood-brain barrier-penetrating small-molecule inhibitor that selectively inhibits the cyclin-dependent kinases CDK2, CDK4, and CDK6. GDC-4198 exerts its antiproliferative effects by inhibiting the phosphorylation of retinoblastoma protein (pRb) through the disruption of cyclin-CDK complex activity, thereby interfering with the cell cycle. GDC-4198 shows potential for application in the treatment of tumors such as hormone receptor-positive/HER2-negative breast cancer, lung cancer, and colorectal cancer.
