Brigimadlin (BI 907828) is a potent, orally bioavailable MDM2-p53 antagonist that disrupts the protein-protein interaction to reactivate p53 tumor suppressor function. It has demonstrated antitumor activity in preclinical research models, including studies of metastatic biliary tract cancer (BTC), pancreatic ductal adenocarcinoma (PDAC), lung adenocarcinoma, and bladder cancer, both in vitro and in vivo.

Bioactivity

• In Vitro:
  Brigimadlin (at concentrations ranging from 0-10 nM; over 48 hours) inhibits the proliferation of BT48, BT50, BT67, BT69, BT89, and BT94, with respective IC50 values of 58.5, 21.1, 37.9, 89.8, 16.7, and 46.8 pM.
