Flow-cytometry using anti-CD48 antibody OX-45 Staining of rat (Rattus norvegicus) lymphocytes with either an isotype control (A) or the rabbit-chimeric version of OX-45 (B, orb2645696) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using an Alexa Fluor ® 488 conjugated donkey anti-rabbit antibody and cells analysed on a FACSCanto flow-cytometer.

Western Blot using anti-CD48 antibody OX-45 Rat spleen (A), thymus (B) and lymph node (C) tissue lysate samples (35 µg protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of OX-45 (orb2645696) at 0.001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for CD48 is ~37-56kDa and it is heavily glycosylated as well as lipidated (UniProt ID: P10252). orb2645696 successfully detected CD48 in rat spleen, thymus and lymph node tissue lysates.
