Immunofluorescence staining of fixed mouse thymocytes with anti-CD48 antibody OX-78. Immunofluorescence analysis of paraformaldehyde fixed mouse (Mus musculus) thymocytes on Shi-fix™ coverslips, permeabilized with 0.15% Triton stained with the chimeric rabbit version of OX-78 (orb2645691) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (1 µg/ml), showing membrane staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom orb2645691, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor® 488 secondary antibody.

Western Blot using anti-CD48 antibody OX-78 Mouse spleen (A), thymus (B) and lymph node (C) tissue lysate samples (35 µg protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of OX-78 (orb2645691) at 0.0001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for CD48 is ~37-56kDa and it is heavily glycosylated, as well as lipidated (UniProt ID: P18181). orb2645691 successfully detected CD48 in mouse spleen, thymus and lymph node tissue lysates.
