Description

NS-1643 is a Human ether-a-go-go related gene (hERG) KV11.1 channel activator (EC50 = 10.5 μM). NS1643 increased both steady-state and tail current at all voltages tested. The EC(50) value for HERG channel activation was 10.5 microM. HERG channel activation by small molecules such as NS1643 increases the repolarization reserve and presents an interesting new antiarrhythmic approach.

Bioactivity

• In Vivo:
  Animal model: NSG mice with human-derived TNBC tumor xenografts. Dosage: 6 mg/kg. Administration: Intraperitoneal injection; 6 mg/kg; twice per week. Result: Significantly reduceed tumor. growth and the metastatic liver tumors were significantly smaller than those in the control group. Decreased levels of human nuclear antigen (HNA).
• In Vitro:
  Western blot analysis. Cell line: MDA-MB-231, SKBR3 and MCF7 breast cancer cell lines. Concentration: 50 μM. Incubation time: 24 h. Result: Decreased Vimentin, N-cadherin and CD44 levels, and increased E-cadherin in breast cancer cell lines.
