HighYield T7 PURE RNA Synthesis Kit is designed to produce large amounts of RNA via in vitro transcription with reduced double-stranded RNA (dsRNA) formation using a modified T7 RNA Polymerase (T7 PURE).\ndsRNA is a critical transcriptonal by-product of wildtype T7 RNA known to trigger an innate immune response and reduction of dsRNA levels requires time-consuming multi-step chromatographic purifcation [1-4]\nHighYield T7 PURE RNA Synthesis Kit is therefore ideally suited for the preparation of RNA probes were the dsRNA impurity level is critical for the subsequent application (e.g. mRNA preparation for cell culture experiments). It reduces down-stream purification efforts to reduce/remove dsRNA contamination.\nOne kit contains sufficient reagents for 50 reactions of 20 μl each (7.5 mM each NTP). A 20 μl reaction yields about 100-120 μg RNA after 30 min and 140-160 μg RNA after 2h incubation (1 μg T7 control template, 1.4 kb RNA transcript). Yields may however vary depending on the template (promoter design, sequence length, secondary structure formation).
