SynuClean-D (SC-D) is a small molecule inhibitor of α-synuclein aggregation. It blocks fibril propagation and disassembles pre-formed fibrils, demonstrating efficacy in both in vitro assays and in vivo models of Parkinson's disease by protecting dopaminergic neurons from degeneration.

Bioactivity

• In Vivo:
  SynuClean-D-treated worms show decreased α-synuclein aggregation in muscle and a concomitant motility recovery. SynuClean-D is able to rescue dopaminergic neurons from α-synuclein-induced degeneration. In two parkinson’s disease (PD) Caenorhabditis elegans models, SynuClean-D expresses α-synuclein either in muscle or in dopaminergic neurons and significantly reduces the toxicity exerted by α-synuclein.
• In Vitro:
  SynuClean-D prevents fibril propagation in protein-misfolding cyclic amplification assays and decreases the number of α-synuclein inclusions in human neuroglioma cells. SynuClean-D significantly reduces the aggregation of wild-type α-synuclein and the familiar A30P and H50Q variants in a substoichiometric molar ratio. SynuClean-D can bind to cavities in mature α-synuclein fibrils.
