Application notes | This antibody can bind human glypican 3 expressed on the surface of 293S cells, HepG2 X1 cells and JHH7 cells in a flow cytometric assay. The binding characterization of this antibody to huGPC3, cynoGPC3, and HepG2 cells was done using ELISA and immunohistochemistry. This antibody strongly reacts with hepatic cancer tissue microarray, JHH cells, HepG2 cells, and cells stably transfected with GPC3 in immunohistochemistry. This antibody also detects human, cynomolgus monkey, rat, and mouse GPC3 by western blot. This antibody was also used in the generation of an antibody drug conjugate with maleimide acetal PNU-159682 or monomethyl disulfide N10-linked PBD. The ADCC activity of the antibody drug conjugate was investigated using a human HepG2 X1 and JHH7 xenograft model and substantial tumor growth inhibition was achieved with 7H1-disulfide-PBD (US9926377). |
