• Immunogen: This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region of human Wnt1 protein.
• Purity: This product was affinity purified from monospecific antiserum by immunoaffinity chromatography. This antibody reacts with human and mouse Wnt1 protein. A BLAST analysis was used to suggest cross-reactivity with Wnt1 from mouse, human, rat, bovine, dog, macaque, and opossum based on a 100% homology with the immunizing sequence. Partial cross-reactivity is expected against chicken Wnt1 based on a 91% sequence homology. Cross-reactivity with Wnt1 from other sources has not been determined.

Expression of Wnt1 in kidney homogenates of sham and nephrectomized rats. The expression of Wnt1 in kidney homogenates of sham and nephrectomized rats 18 weeks after the surgery was analyzed by immunoblotting. Blotting of GDPH served as loading control. Results are derived from 4-5 animals per group.

Western blot using Biorbyt's affinity purified anti-Wnt1 antibody shows detection of endogenous Wnt1. Lane 1: human-derived MCF7 cell lysate (p/n orb348664). Lane 2: mouse-derived 3T3 cell lysate (p/n orb348714). The band at ~41kDa, indicated by the arrowhead, corresponds to Wnt1. After transfer, the membrane was blocked with 5% BLOTTO (p/n orb348624). Primary antibody was used at a 1:1400 dilution in PBS containing 1% BLOTTO. The specificity of the antibody was confirmed by peptide competition which completely blocked reaction of the antibody with Wnt1 (data not shown).
