ELISA of anti-VISTA antibody on VISTA-Fc fusion protein. Binding curve of the rmeric version of the anti-VISTA antibody MH5A (orb1671801; blue line) to an ELISA plate coated with mouse VISTA-Fc fusion protein (Pr00164-1.9) at a concentration of 5 ug/ml. A 3-fold serial dilution from 10000 to 1.7 ng/ml was performed using orb1671801. For signal detection- a 1:4000 dilution of anti-r1 HRP antibody was used.
Immunofluorescence staining of fixed mouse splenocytes with anti-PD-1H antibody MH5A. Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes permewith 0.15% Triton and stained with the chimeric mouse IgG1 version of MH5A (orb1671801) at 10 ug/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 ug/ml)- showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right- top-bottom orb1671801- DAPI- merged channels and an isotype control. The isotype control was stained with an anti-unknown specificity antibody (0) followed by Alexa Fluor® 488 secondary antibody.
