  This protein A purified antibody has been tested for use in ELISA and western blot. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 42 kDa in size corresponding to RNF34 protein by western blotting in the appropriate tissue or cell lysate or extract. Isoforms 1 and 2, which are equivalent in size, should both cross-react with this antibody.

• Immunogen: This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a recombinant protein corresponding to amino acids 1-373 of human RNF34 protein.
• Purity: This protein A purified antibody is directed against human RNF34 protein. The product was purified from monospecific antiserum by protein A chromatography followed by exhaustive dialysis against the buffer stated above. A BLAST analysis was used to suggest cross-reactivity with RNF34 protein from orangutan (99%), bovine (91%), dog (89%), mouse (88%) and rat (85%) based on a high degree of protein:protein homology with the immunizing sequence. Expect reactivity with isoform 1 and 2 of RNF34. Reactivity against homologues from other sources is not known.

• Storage: Store vial at 4° C prior to restoration. For extended storage aliquot contents and freeze at -20° C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.

Western blot using Biorbyt's Protein A Purified anti-RNF34 antibody shows detection of human RNF34 (arrowhead) in lysate. Lanes corresponding to empty vector 293T cell lysate (mock, left); RNF34 transfected lysate (middle) and RFFL transfected lysate (right), are shown using 20 µl of lysate per lane. Lysates were prepared from equivalent numbers of cells. Data presented demonstrate that this reagent is specific for RNF34. After SDS-PAGE and transfer, the membrane was probed with the primary antibody diluted to 1:1000 using 5% BLOTTO, 0.1% Tween-20 in PBS as the diluent. Incubation occurred for 1 h at room temperature.
