In Vivo
METHODS: To investigate the effect of Pioglitazone on insulin resistance, Pioglitazone (10 and 30 mg/kg) was orally administered to ob/ob and adipo-/-ob/ob mice once daily for 14 days. RESULTS: Pioglitazone improves insulin resistance and diabetes, which may be lipocalin-dependent in the liver but not in skeletal muscle. METHODS: To investigate the effect of Pioglitazone on cardiac remodeling, diabetic nephropathy rats were treated with oral administration of Pioglitazone (10 mg/kg) once daily for 4 weeks. RESULTS: Pioglitazone significantly reduced body weight (BW), cardiac hypertrophy, elevated blood glucose levels, and related dyslipidemia.
In Vitro
METHODS: HT-1080, MDA-MB-231, and PC-3 cells were treated with Pioglitazone for 3 days, and target cell toxicity was measured using MTT assay. RESULTS: Pioglitazone did not affect cell growth at 100 μM.
