Storage & Handling
−| Storage | Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing. |

Western Blot of Anti-OspA Antibody. Lane 1: Opal Pre-stained Molecular Weight Marker. Lane 2: 10 µg HeLa spiked 50 ng OspA. Lane 3: 10 µg HeLa spiked 20 ng OspA. Lane 4: 10 µg HeLa spiked 10 ng OspA. Lane 5: 10 µg HeLa Whole Cell Lysate (p/n orb348668). Lane 6: 10 µg HeLa spiked 50 ng OspB. Lane 7: 10 µg HeLa spiked 50 ng OspC. Primary Antibody: Anti-OspA at 1:1000 overnight at 2-8°C. Secondary Antibody: Goat Anti-Rabbit IgG HRP (p/n orb347654) at 1:70000 for 30 min at RT. Block: BlockOut Buffer (p/n orb348644). Predicted MW: ~28.1kDa OspA protein, 70.5kDa MBP-OspA fusion. Observed MW: ~30kDa, ~75kDa.

Western blot showing detection of 0.1 µg of recombinant OspA protein. Lane 1: Molecular weight markers. Lane 2: MBP-OspA fusion protein (arrow; expected MW: 70.5 kDa). Lane 3: MBP alone. Protein was run on a 4-20% gel, then transferred to 0.45 µm nitrocellulose. After blocking with 1% BSA-TTBS (p/n orb348540, diluted to 1X) overnight at 4°C, primary antibody was used at 1:1000 at room temperature for 30 min. HRP-conjugated Goat-Anti-Rabbit (p/n orb347654) secondary antibody was used at 1:40000 in orb348637 blocking buffer.
