DescriptionFlaviviruses belong to the family of flaviviridae, most of which are arboviruses mainly including Zika virus (ZIKA), Yellow fever virus (YFV), West Nile virus (WNV) and Japanese Encephalitis Virus (JEV).Flaviviruses are small, enveloped viruses with a positive-strand RNA genome, which translates into three structural proteins and seven non-structural proteins. NS1, one of the non-structural proteins, is a highly conserved dimer protein with the molecular weight from 46~55 kDa depending on the extent of glycosylation. The glycosylation of NS1 plays a role in efficient secretion, virulence and viral replication. NS1 exists as a monomer, a dimer (membranebound protein, mNS1) and a hexamer (secreted protein, sNS1). The NS1 inside cells is crucial to viral replication, while the sNS1 and mNS1 have been reported to elicit the immune response. All flavivirus NS1 genes share a high degree of homology and are 1056 nucleotides in length, encoding a 352-amino-acid polypeptide. The conserved regions of NS1 exist in various flaviviruses. The increasing concentration of NS1 directly correlates with disease severity and incresed viremia. The NS1 protein is an esssential target for inhibitor design. The secreted and cell surface-associated NS1 are highly immunogenic. The NS1 is an important biomarker for early diagnosis of the flaviviral infections.
