Flow cytometry using the Anti-Tim-3 antibody 2C12. Paraformaldehyde fixed mouse splenocytes permewith 0.5% Triton were stained with anti-unknown specificity antibody (3.0; isotype control - black line) or the r version of 2C12 (orb1671768 - blue line) at a dilution of 1:100 for 1h at RT. After washing- the bound antibody was detected using a goat anti-r AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
Immunofluorescence staining of fixed RAW264.7 cells with anti-Tim-3 antibody 2C12. Immunofluorescence analysis of paraformaldehyde fixed RAW264.7 cells on Shi-fix™ coverslips stained with the chimeric r version of 2C12 (orb1671768) at 10 ug/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 ug/ml)- showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right- top-bottom orb1671768- DAPI- merged channels and an isotype control. The isotype control was an unknown specificity antibody (.1) followed by staining with Alexa Fluor® 488 secondary antibody.
ELISA of anti-TIM-3 antibody on TIM-3-Fc fusion protein. Binding curves of the rmeric version of the anti-TIM-3 antibody 2C12 (orb1671768; blue line) and isotype control (3.0- anti-Fluorescein; red line) to an ELISA plate coated with mouse TIM-3-Fc fusion protein (Pr00228-1.9) at a concentration of 5 ug/ml. A 3-fold serial dilution from 3000 to 0.0169 ng/ml was performed using orb1671768. For signal detection- anti-r1 HRP antibody was used.
