Draining lymph nodes were fused with Sp2/0-Ag14 cells, selected in HAT (hypoxanthine/aminopterin/thymidine) medium and supernatants were screened for specificity by ELISA and flow cytometry using TIGIT-transfectants.

ELISA of anti-TIGIT antibody on TIGIT-Fc fusion protein. Binding curves of the rmeric IgG version of the anti-TIGIT antibody 4D4 (orb1671807; red line) and isotype control (3.0- anti-Fluorescein; blue line) to an ELISA plate coated with mouse TIGIT-Fc fusion protein (Pr00163-1.9) at a concentration of 5 ug/ml. A 3-fold serial dilution from 10-000 to 0.17 ng/ml was performed using orb1671807. For signal detection- a 1:4000 dilution of HRP-lnti-r1 antibody was used. The second increase in e at 1000 ng/ml is s uggestive of a possible secondary binding epitope for orb1671807 on Pr00163-1.9.
