    ImmunogenThis antibody was prepared from whole rabbit serum produced by repeated immunizations with a polyhistidine-tagged TTFC fusion protein expressed in E. coli corresponding to full length TTFC. The polyhistidine-tag was placed at the amino terminus of the protein. The recombinant protein was purified from soluble bacterial extracts by metal-affinity and ion-exchange chromatography.

    Dilution rangeThis antibody has been tested for use in ELISA and western blot. Specific conditions for reactivity should be optimized by the end user. For western blot experiments, one can expect a band of ≈52 kDa in size corresponding to full-length TTFC. For direct ELISA, coat plates with 1 μg of recombinant TTFC (0.1ml/well). Antigen-specific signal can be detected at a dilution up to 1:10,000,000. For antigen capture ELISA, 0.1 ml of the diluted antisera (1:4000) was able to detect 6 picograms of TTFC using monoclonal mouse anti-TTFC as the antigen-detection antibody (0.5 μg/ml, Roche Biosciences). Sera diluted up to 100,000-fold can easily detect 600 picograms of recombinant TTFC by Western blot processed with ECL Plus Western Blotting and Detection System (Amersham Biosciences, 1 sec film exposure).
