Description

Pyrogallol red (PGR) is a polyphenolic dye that serves as a highly reactive probe for detecting reactive oxygen and nitrogen species. It is rapidly bleached by superoxide anions with a 1:2 stoichiometry (one PGR molecule consumed per two superoxide radicals), while remaining insensitive to hydrogen peroxide. PGR also reacts efficiently with hypochlorite (0.7 molecules bleached per hypochlorite) and carbonate radicals, with initial consumption rates reaching 24.6 µM min⁻¹ from SOD1 peroxidase activity. Commonly employed in microplate-based spectrophotometric assays, PGR is used to quantify superoxide release from NADPH oxidase, measure redox cycling of nitro compounds, and evaluate antioxidant capacities.
