E. coli DNA Ligase is a DNA ligase that catalyzes the joining of sticky ends of double-stranded DNA or the repair of cuts in double-stranded DNA. The enzyme catalyzes the formation of a phosphodiester bond between the 5’phosphate and 3’hydroxyl groups in double-stranded DNA, and the ligation reaction requires NAD (nicotinamide adenine dinucleotide) as a coenzyme. E. coli The DNA ligase was active within a certain temperature range (4~37℃) and lost its activity after incubation at 65℃ for 20 min.E. coli DNA Ligase is more specific for substrate selection than T4 DNA Ligase, and is recommended if only ligation of sticky ends is desired and inhibition of ligation of flat ends or RNA molecules is desired.
