  To prepare the PIPES buffer, dissolve 173 g of 1,4-piperazinediethanesulfonic acid in 1 L of deionized water. Adjust the pH of the solution to 6.8 using sodium hydroxide pellets. This buffer is efficacious in mitigating lipid loss and artifacts induced by glutaraldehyde fixation .
