Description | Rabbit polyclonal to PERFORIN.PERFORIN (PRF1) is a cytolytic protein found in the granules of CD8 T-cells and NK cells. Upondegranulation, PERFORIN inserts itself into the target cell's plasma membrane, forming a pore. Thelytic membrane-inserting part of PERFORIN is the MACPF domain, which shares homology withcholesterol-dependent cytolysins from Gram-positive bacteria. PERFORIN has structural andfunctional similarities to complement component 9 (C9). Like C9, PERFORIN creates transmembranetubules and is capable of lysing non-specifically a variety of target cells. PERFORIN is one of themain cytolytic proteins of cytolytic granules, and it is known to be a key effector molecule for T-cellandnatural killer-cell-mediated cytolysis. Defects in PERFORIN cause familial hemophagocyticlymphohistiocytosis type 2 (HPLH2), a rare and lethal autosomal recessive disorder of early childhood. |
