The MitoPT TMRM Mitochondrial Depolarization Assay Kit detects mitochondrial membrane depolarization using the fluorescent dye TMRM. In healthy mitochondria with a negative charge due to polarization, TMRM accumulates and emits orange fluorescence. However, when the mitochondrial membrane potential collapses in apoptotic or metabolically stressed cells, the TMRM reagent disperses throughout the cell cytosol, resulting in a dramatic drop in fluorescence levels.
