Purity | This antiserum is directed against the extracellular domain of the human IL-1R type II, but precipitates the intact receptor expressed on human cells. In general, this antibody also detects primate IL-1RII. The antibody does not recognize human IL-1, IL-1Ra or IL-1R type I. In ELISA formats and other immunoreactive assays, this antibody will detect the extracellular domain of the IL-1R type II (also called soluble IL-1R type II) found in human body fluids, particularly in the circulation, urine and supernatants of cells and synovial fluid. |

Storage | Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing. |

Application notes | This antiserum against Human IL-1 Receptor Type II (IL-1RII) has been tested by western blot and is suitable for use in ELISA, radioimmunoassays and immunoprecipitation. Reactivity in other immunoassays is unknown. IL-1RII has been termed p68 or CD121 in some laboratories. IL-1RII has been shown to bind IL-1, IL-1a and IL-1b. |

Western blot using Biorbyt's anti-IL-1RII antibody. This blot shows detection of an IL-1RII-GST fusion protein (~500 ng, lane 1, green, 800 nm channel). Minimal reactivity is observed against GST (data not shown). Protein was resolved on a 4-20% Tris-Glycine gel by SDS-PAGE and transferred onto nitrocellulose. After blocking, the membrane was probed with the primary antibody diluted to 1:1000. Incubation was for 2 hrs at room temperature followed by washes and reaction with a 1:10000 dilution of IRDye® 800 conjugated Gt-a-Rabbit IgG (H&L) MX10 for 45 min at room temperature. Molecular weight markers are shown for size comparison (lane M, red, 700 nm channel).
