HighYield T7 P&L RNA Synthesis Kit is designed to produce large amounts of RNA via in vitro transcription with a modified T7 RNA Polymerase. Proline 266 has been replaced by leucine (P266L) which has been associated with: decreased abortive transcription. increased 5' homogeneity of transcripts synthesized from A-initiating phi2.5 promoter; increased 5' incorporation efficiency of GTP analogs The resulting RNA can subsequently be used for a variety of appli-cations such as microinjection, transfection or in vitro translation experiments, structure/function studies or probes for in situ hy-bridization and Northern Blot experiments. One kit contains sufficient reagents for 50 reactions of 20 µl each (7.5 mM each NTP). A 20 µl reaction yields about 140-160 µg RNA after 30 min incubation (1 µg T7 control template, 1.4 kb RNA transcript). Yields may however vary depending on the template (promoter design, sequence length, secondary structure formation).
